In the Civil Survey of 1654, it is estimated to be around 60 cottages in and around the castle at Nenagh. The population was 275 persons in 1657. This number is unlikely to have increased significantly by the time of the castle's dismantling in 1692. By 1841 the population had increased to 8618. A century or so later this had decreased to 4517 by 1946. It has had a population of 9,895 as of the Census of 2022.

In 1760 the main thoroughfare through the town was entered at the Toll Booth at Annebrook where it took a sharp right which led across to Smithville (Yewston) and came down Bournes Lane entering onto Silver St. This road then continued down past the Market House at the Market Cross, through Castle St before turning right down Chapel lane, past the Franciscan Friary and entering on the Dublin Road at the Spout. From there the road continued on out past Riverston where the River had to be forded, Bennets Bridge not yet having been built.
